To understand how the spikes affect virion mobility and infectivity, we investigate the diffusivity of spike-decorated structures using mesoscopic-hydrodynamic simulations. Furthermore, we explored the interplay between affinity and passive viral transport. Our results revealed that the diffusional mechanism of SARS-CoV-2 is strongly influenced by the size and distribution of its spikes. We propose and validate a universal mechanism to explain the link between optimal virion structure and maximal infectivity for many virus families.Neonatal head circumference (HC) not only represents the brain size of Homo sapiens, but is also an important health risk indicator. Addressing a lack of comparative studies on head size and its variability in term and preterm neonates from different populations, we aimed to examine neonatal HC by gestation according to a regional reference and a global standard. Retrospective analysis of data on neonatal HC obtained from the Lithuanian Medical Birth Register from 2001 to 2015 (423 999 newborns of 24-42 gestational weeks). The varying distribution by gestation and sex was estimated using GAMLSS, and the results were compared with the INTERGROWTH-21st standard. Mean HC increased with gestation in both sexes, while its fractional variability fell. The 3rd percentile matched that for INTERGROWTH-21st at all gestations, while the 50th and 97th percentiles were similar up to 27 weeks, but a full channel width higher than INTERGROWTH-21st at term. INTERGROWTH-21st facilitates the evaluation of neonatal HC in early gestations, while in later gestations, the specific features of neonatal HC of a particular population tend to be more precisely represented by regional references.

The rapid emergence and expansion of novel SARS-CoV-2 variants threatens our ability to achieve herd immunity for COVID-19. These novel SARS-CoV-2 variants often harbor multiple point mutations, conferring one or more evolutionarily advantageous traits, such as increased transmissibility, immune evasion and longer infection duration. In a number of cases, variant emergence has been linked to long-term infections in individuals who were either immunocompromised or treated with convalescent plasma. In this paper, we used a stochastic evolutionary modeling framework to explore the emergence of fitter variants of SARS-CoV-2 during long-term infections. We found that increased viral load and infection duration favor emergence of such variants. While the overall probability of emergence and subsequent transmission from any given infection is low, on a population level these events occur fairly frequently. Targeting these low-probability stochastic events that lead to the establishment of novel advantageous viral variants might allow us to slow the rate at which they emerge in the patient population, and prevent them from spreading deterministically due to natural selection. Our work thus suggests practical ways to achieve control of long-term SARS-CoV-2 infections, which will be critical for slowing the rate of viral evolution.The pandemic of SARSCov2 infection has created a challenge in health services worldwide. The HACOR scale and the ROX index have a moderate predictive capacity to predict failures to the HFNC strategy. They can be used in conjunction with other clinical variables to define which patients may require invasive mechanical ventilation.We aimed to identify distinct longitudinal trends of LDL-cholesterol (LDL-C) levels and investigate these trajectories' association with statin treatment. This retrospective cohort study used electronic health records from 8592 type 2 diabetes patients in North Karelia, Finland, comprising all primary and specialised care visits 2011‒2017. We compared LDL-C trajectory groups assessing LDL-C treatment target achievement and changes in statin treatment intensity. Using a growth mixture model, we identified four LDL-C trajectory groups. The majority (85.9%) had "moderate-stable" LDL-C levels around 2.3 mmol/L. The second-largest group (7.7%) consisted of predominantly untreated patients with alarmingly "high-stable" LDL-C levels around 3.9 mmol/L. The "decreasing" group (3.8%) was characterised by large improvements in initially very high LDL-C levels, along with the highest statin treatment intensification rates, while among patients with "increasing" LDL-C (2.5%), statin treatment declined drastically. In all the trajectory groups, women had significantly higher average LDL-C levels and received less frequent any statin treatment and high-intensity treatment than men. Overall, 41.9% of patients had no statin prescribed at the end of follow-up. Efforts to control LDL-C should be increased-especially in patients with continuously elevated levels-by initiating and intensifying statin treatment earlier and re-initiating the treatment after discontinuation if possible.The aim of this study was to assess the association between dietary inflammatory index (DII) and non-invasive markers of liver status in adults. This cross-sectional study was performed on 8520 adults, recruited in Ravansar Non-Communicable Diseases (RaNCD) cohort study, western Iran. The DII score was calculated based on participants' dietary intakes obtained from Food Frequency Questionnaire (FFQ). Fatty Liver Index (FLI) score was calculated by anthropometric measurements and some non-invasive markers of liver status. Linear regression models were applied to estimate the associations and adjust the possible confounding factors. A greater DII score was significantly associated with higher energy intake, body mass index (BMI), body fat mass (BFM), blood pressure, and FLI (P  less then  0.001). Participants with the highest DII score had a significantly higher consumption saturated fat, trans fat and red meat than those in the lowest quartile (P  less then  0.001). After adjustments of age and sex, participants in the highest quartile of the DII score had a greater risk of FLI (β 0.742, 95% CI 0.254, 0.601). More pro-inflammatory diet in participants was associated with a higher FLI. The DII score was positively associated with non-invasive liver markers. Thus, having an anti-inflammatory diet can help balance liver enzymes, reduce obesity, and decrease fatty liver.When laser beams propagate through media with non-vanishing absorption, the media is heated resulting in a change of the refractive index, which can lead to thermal lensing and thermal blooming. However, experimental details about both phenomena for propagations in water are lacking, especially for high-power lasers in the kilowatt range. We show that significant thermal lensing occurs only for high input powers before the onset of convective flow, while for low input powers, no strong thermal lens arises. After the onset of water flow, thermal blooming occurs at low input powers comparable to that known for propagations over kilometres in the air. However, for high input powers a thermal blooming on a qualitatively higher level is shown. By wavefront sensing, the change of refractive index distribution in water is investigated. This clearly shows the fast development of a strong thermal lens for high input powers and the onset of convection. Furthermore, a qualitatively good agreement of the accompanying simulations is observed. It is found that the absorption coefficient is linear with a value of [Formula see text] at least up to 7.5 kW, i.e. 8 [Formula see text]. For the evaluation of PSMA expression levels, male NOD/SCID mice bearing prostate cancer (PCa) xenografts of C4-2 (PSMA+++), 22Rv1 (PSMA+) and PC-3 (PSMA-) were administered [18F]AlF-PSMA-11 with a medium MAapp (20.24 ± 3.22 MBq/nmol). SUVmean and SUVmax values were respectively 3.22 and 3.17 times higher for the high versus low PSMA expressing tumors (p  less then  0.0001). To evaluate the effect of varying MAapp, C4-2 and 22Rv1 xenograft bearing mice underwent additional [18F]AlF-PSMA-11 imaging with a high (211.2 ± 38.9 MBq/nmol) and/or low MAapp (1.92 ± 0.27 MBq/nmol). SUV values showed a significantly increasing trend with higher MAapp. Significant changes were found for SUVmean and SUVmax between the high versus low MAapp and medium versus low MAapp (both p  less then  0.05), but not between the high versus medium MAapp (p = 0.055 and 0.25, respectively). The effect of varying MAapp was more pronounced in low expressing tumors and PSMA expressing tissues (e.g. salivary glands and kidneys). Overall, administration of a high MAapp increases the detection of low expression tumors while also increasing uptake in PSMA expressing tissues, possibly leading to false positive findings. In radioligand therapy, a medium MAapp could reduce radiation exposure to dose-limiting organs with only limited effect on radionuclide accumulation in the tumor.Tandem paired nicking (TPN) is a method of genome editing that enables precise and relatively efficient targeted knock-in without appreciable restraint by p53-mediated DNA damage response. TPN is initiated by introducing two site-specific nicks on the same DNA strand using Cas9 nickases in such a way that the nicks encompass the knock-in site and are located within a homologous region between a donor DNA and the genome. This nicking design results in the creation of two nicks on the donor DNA and two in the genome, leading to relatively efficient homology-directed recombination between these DNA fragments. In this study, we sought to identify the optimal design of TPN experiments that would improve the efficiency of targeted knock-in, using multiple reporter systems based on exogenous and endogenous genes. We found that efficient targeted knock-in via TPN is supported by the use of 1700-2000-bp donor DNAs, exactly 20-nt-long spacers predicted to be efficient in on-target cleavage, and tandem-paired Cas9 nickases nicking at positions close to each other. These findings will help establish a methodology for efficient and precise targeted knock-in based on TPN, which could broaden the applicability of targeted knock-in to various fields of life science.Postpartum depression is an important mental health issue not only for the mother but also for the child's development, other family members, and the society. Behavioral experiments to investigate postpartum depression-like behaviors should be conducted without stress because behavioral tests affect rearing behaviors such as lactation. Therefore, we developed a new mouse model of postpartum depression using a psychological stress method. Mating partners were made to witness their partners experiencing social defeat stress and then listen to their cries. Emotional stress loading during pregnancy significantly increased postpartum depression-like behaviors. Postpartum depression also affected nurturing behaviors and caused disturbances in pup care. Furthermore, nesting behavior was impaired in the stressed group, suggesting that the observation of nesting behavior may be useful for assessing social dysfunction in postpartum depression. In total, 53 diseased fruit samples were sampled from eight types of fruit trees (jackfruit, rose apple, mangosteen, plum, snake fruit, rambutan, strawberry, and avocado). Based on morphological characteristics, 44 isolates were classified as belonging to pestalotioid taxa. Of these isolates, seven with distinct characteristics were selected for identification using molecular analysis, and six isolates were identified as Neopestalotiopsis and one as Pseudopestalotiopsis. In the cross-inoculation experiment, the isolates exhibited nonhost specificity and could infect at least two host plants. The isolates were used to screen for a potential biocontrol resource using six crude plant extracts (clove, ginger, lemongrass, mangosteen, roselle, and turmeric).

Objectives As one of the most serious malignant carcinomas that threaten the life of sufferers constantly, gastric cancer has attracted a lot of interest among researchers. miR-34a, a member of hundreds of microRNAs (miRNAs), has been elucidated to exert a suppressive role in gastric cancer tumorgenesis based on previous extensive studies. Our study was performed with the aim to explore the functional effects of miR-34a and its predictive target programmed death ligand 1 (PDL1) in gastric cancer development. Methods We employed reverse transcription-quantitative polymerase chain reaction (RT-qPCR) and Western Blot analysis to investigate the regulatory effect of miR-34a on PDL1 mRNA and the corresponding protein expression. The CCK-8 and colony formation assays were used to validate the influence of the combination of miR-34a and PDL1 on the proliferation of gastric tumor cells. Meanwhile, the migration and invasion of gastric tumor cells were measured using Transwell assay. Results PDL1 was targeted and negatively modulated by miR-34a. In addition, the re-expression of miR-34a suppressed the proliferation as well as the migration and invasion of gastric tumor cells, whereas PDL1 reduced the aforementioned inhibitory effect. Conclusions PDL1 is the downstream gene of miR-34a, which can act as an anti-oncogene in gastric cancer. The miR-34a/PDL1 axis might provide a promising anticancer therapeutic approach for the clinical diagnosis, treatment, and prognosis of gastric cancer.Pyridoxal 5'-phosphate (PLP) is an essential cofactor that participates in ∼4% enzymatic activities cataloged by the Enzyme Commission. The pdxST genes from Bacillus subtilis 168 were introduced into the tyrosine phenol-lyase (TPL)-overexpressing Escherichia coli BL21(DE3) strain. TPL and PdxST were co-expressed with a double-promoter or a compatible double-plasmid system. The 3,4-dihydroxyphenylacetate-L-alanine (L-DOPA) titer did not increase with the increase in the intracellular PLP concentration in these strains with TPL and PdxST co-expression. Therefore, it is necessary to optimize the intracellular PLP metabolism level so as to achieve a higher L-DOPA titer and avoid the formation of L-DOPA-PLP cyclic adducts. Curcumin, as a known polyphenolic compound extracted from turmeric, is a chemotherapeutic used in some cancer treatments in china. However, the effects of curcumin on the survivability of GB cells remain to be elucidated. Methods We performed a CCK8 assay to detect viability of GB cells following treatments with curcumin and examined the migration and invasion ability of these cells using the wound-healing and transwell invasion assays. The cell proliferation and apoptotic proteins were detected by Western blot analyses. We detected that curcumin decreased p-AKT and p-mTOR protein expression, and promoted apoptosis of U251 and U87 GB cells. In addition, we administered curcumin to nude mice and found that curcumin decreased the tumor volume, caused necrosis of tumor tissue, and significantly enhanced the PTEN and p53 expression in vivo. Conclusions These results indicated that curcumin inhibited proliferation by decreasing the p-AKT/p-mTOR pathway, and promoted apoptosis by increasing the PTEN and p53 expression. Despite the robust nature of category-advantage in color perception, the related cognitive and neural mechanisms are not fully understood. Some views attribute this effect to early alteration of visual processing of color while others attribute it to post-perceptual conceptual processing. The current study investigated the causal role of the left anterior temporal lobe (ATL), as a post-perceptual semantic hub, in categorical color perception. We modulated the activity of the left ATL via cathodal tDCS or sham stimulation (within-subject) while participants were discriminating between successive presentations of color patches. Without stimulation, we found a category-advantage effect in both accuracy and response times. The inhibition of left ATL eliminated the category-advantage effect in terms of RTs but not accuracies. Our results point at the causal role of ATL in categorical color perception and provide indirect support for a post-perceptual processing account of this robust phenomenon.SARS-CoV-2-related SARIs (severe acute respiratory infections) have a major impact on public health; moreover, healthcare workers (HCWs) are exposed to a high biological risk. The aim of this study was to show the prevention procedures in place in the University Hospital of Bari (Apulia Region, Southern Italy) to reduce this risk to HCWs, consisting of the enhancement of preventive measures and the activation of a report system to collect HCWs' contacts. To date, 23 confirmed cases of infections (0.4% of all HCWs) have been reported in a 30-day observation period. These results show that correct management of HCWs' contacts is essential to avoid nosocomial clusters.

Whilst significant advances have been made in TB diagnostics, there is still a need to optimize the diagnostic yield of Xpert MTB/RIF and Xpert Ultra in EPTB samples. Research is needed to facilitate standardization and optimal preparation of samples as well as understanding the role of Xpert MTB/RIF and Xpert Ultra in different burden settings. Alongside the current GeneXpert platform, the launch of rapid second-line drug resistance polymerase chain reactions and whole genome sequencing may help tackle the global health burden with a more comprehensive diagnostic approach and appropriate treatment.
Orbital veins such as the retinal veins and episcleral veins drain into the cavernous sinus, an intracranial venous structure. We studied the effects of acute intracranial pressure (ICP) elevation on episcleral venous pressure, intraocular pressure and retinal vein diameter in an established non-survival pig model.

In six adult female domestic pigs, we increased ICP in 5 mm Hg increments using saline infusion through a lumbar drain. We measured ICP (using parenchymal pressure monitor), intraocular pressure (using pneumatonometer), episcleral venous pressure (using venomanometer), retinal vein diameter (using OCT images) and arterial blood pressure at each stable ICP increment. The average baseline ICP was 5.4 mm Hg (range 1.5-9 mm Hg) and the maximum stable ICP ranged from 18 to 40 mm Hg. Linear mixed models with random intercepts were used to evaluate the effect of acute ICP increase on outcome variables.

With acute ICP elevation, we found loss of retinal venous pulsation and increased episcleral venouive ophthalmic biomarkers to estimate acute ICP elevations following significant neuro-trauma.Diabetic wound infections and pressure ulcers pose a significant challenge to healthcare providers worldwide. The current study provides new and innovative wound care products that reduce inflammation, clear infection, and improve healing in an animal model of pressure ulcers in diabetic rats. Ointment, hydrogel, and nanofiber dressings were synthesized using 5% turmeric, 1% oregano, and 1% chitosan nanoparticles and tested for antibacterial and cytotoxicity in vitro, and wound healing effects in vivo. Turmeric ethanolic extract showed high antioxidant activity compared to Oregano, Chitosan Nanoparticles, and Alginate silver (p-value less then 0.0001). The ointment and hydrogel formulation (5% Turmeric, 1% Oregano, and 1% chitosan) showed lower cytotoxicity compared to the commercial Alginate silver dressing. Ointment, hydrogel formulations, and commercial Alginate silver, showed significant antibacterial activity with 100% efficacy on both Staphylococcus aureus and Escherichia coli (p-value less then 0.0001), compared to nanofibers which showed 50% reduction in bacterial growth (p-value less then 0.0001). The new formulations were tested in a rat model of pressure ulcers. Ointment and nanofibers achieved complete wound healing by day 15 compared to the hydrogel and commercial Alginate silver dressing, which showed higher infection, and the wound remained partially open by day 21. In conclusion, Turmeric, Oregano extracts, and chitosan nanoparticles can be used for effective wound dressings in both diabetic and non-diabetic wounds. At relatively low concentrations, this combination provides a promising new wound treatment formulation that is antibacterial, anti-inflammatory, and antioxidant.Purpose To develop a treatment model of rhegmatogenous retinal detachment (RRD) in which the effects of various vitreous tamponades can be explored. Methods In a primary session, detachment was produced in the right eye of 24 rabbits using vitrectomy, posterior vitreous detachment, retinal break induction, and subretinal injection of viscoelastic solution. The following day, detachments were treated in 16 eyes using SF6 (n = 8) or Healaflow® (HF, a cross-linked hyaluronic acid hydrogel, n = 8) tamponade. Animals were followed for 1 month and thereafter examined macroscopically and morphologically in hematoxylin and eosin-stained sections. Results Retinal detachment (RD) was successfully treated using repeated surgery. We herein describe three patients who developed fatal intracranial hemorrhage (ICH) after carotid artery stenting (CAS).
We retrospectively reviewed 126 patients who underwent CAS from January 2016 to December 2018 and identified 3 patients (2.4%) (all male, mean age of 59 years) who developed ICH after CAS. Two of them developed left basal ganglia hemorrhage with extension into the ventricle and subarachnoid space, and the third patient developed primary ventricular bleeding. One hemorrhage occurred immediately after CAS, whereas the other two occurred 3 hours and 8 hours after the procedure, respectively. The mean stenosis of the treated carotid arteries was 91%. All three hemorrhages were fatal, and the mean time from hemorrhage to death was 50 hours.

ICH is a potentially fatal complication of CAS and often occurs several hours after the procedure. Headache, vomiting, and consciousness disorders are the most common symptoms of ICH. Careful screening to identify high-risk patients and strict management of perioperative blood pressure are important to prevent this complication.
