Rapid and accurate detection of a C-shaped root canal on mandibular second molars can assist dentists in diagnosis and treatment. Oral panoramic radiography is one of the most effective methods of determining the root canal of teeth. There are already some traditional methods based on deep learning to learn the characteristics of C-shaped root canal tooth images. However, previous studies have shown that the accuracy of detecting the C-shaped root canal still needs to be improved. And it is not suitable for implementing these network structures with limited hardware resources. In this paper, a new lightweight convolutional neural network is designed, which combined with receptive field block (RFB) for optimizing feature extraction. In order to optimize the hardware resource requirements of the model, a lightweight, multi-branch, convolutional neural network model was developed in this study. To improve the feature extraction ability of the model for C-shaped root canal tooth images, RFB has been merged with this model. RFB has achieved excellent results in target detection and classification. In the multiscale receptive field block, some small convolution kernels are used to replace the large convolution kernels, which allows the model to extract detailed features and reduce the computational complexity. Finally, the accuracy and area under receiver operating characteristics curve (AUC) values of C-shaped root canals on the image data of our mandibular second molars were 0.9838 and 0.996, respectively. The results show that the deep learning model proposed in this paper is more accurate and has lower computational complexity than many other similar studies. In addition, score-weighted class activation maps (Score-CAM) were generated to localize the internal structure that contributed to the predictions.Idiopathic aplastic anemia (IAA) pathophysiology is dominated by autoreactivity of human leukocyte antigen (HLA)-restricted T-cells against antigens presented by hematopoietic stem and progenitor cells (HSPCs). Expansion of PIGA and HLA class I mutant HSPCs have been linked to immune evasion from T-cell mediated pressures. We hypothesized that in analogy with antitumor immunity, the pathophysiological cascade of immune escape in IAA is initiated by immunoediting pressures and culminates with mechanisms of clonal evolution characterized by hits in immune recognition and response genes. Using a newly implemented bioinformatic framework we found that not only class I but also class II genes were often impaired by acquisition of genetic aberrations. We also demonstrated the presence of novel somatic alterations in immune genes possibly contributing to the evasion from the autoimmune T-cells. Our findings shed light on the mechanisms of immune activation and escape in IAA and define alternative modes of clonal hematopoiesis.The screening of siRNAs targeting 390 human G protein-coupled receptors (GPCRs) was multiplexed in combination with cisplatin against lung cancer cells. While the cell viability measure hardly captured the anticancer effect of siGPCRs, the direct cell count revealed the anticancer potential of diverse GPCRs (46 hits with > twofold growth inhibition, p-value less then 0.01). In combined treatment with cisplatin, siRNAs against five genes (ADRA2A, F2RL3, NPSR1, NPY and TACR3) enhanced the anti-proliferation efficacy on cancer cells and reduced the self-recovery ability of surviving cells after the removal of the combined treatment. Further on-target validation confirmed that the knockdown of TACR3 expression exhibited anticancer efficacy under both single and combined treatment with cisplatin. In this study, we showed that de novo cholesterol biosynthesis induced by ASPP2 depletion in mouse breast cancer cell 4T1 and human breast cancer cell MDA-MB-231 promoted NETs formation in vitro, as well as in lung metastases in mice intravenously injected with ASPP2-deficient 4T1 cells. Simvastatin and berberine (BBR), cholesterol synthesis inhibitors, efficiently blocked ASPP2-depletion induced NETs formation. Cholesterol biosynthesis greatly enhanced Coiled-coil domain containing protein 25 (CCDC25) expression on cancer cells as well as in lung metastases. CCDC25 expression was co-localized with caveolin-1, a lipid raft molecule, and was damped by inhibitor of lipid rafts formation. Our data suggest that cholesterol biosynthesis promotes CCDC25 expression in a lipid raft-dependent manner. Clinically, the expression of CCDC25 was positively correlated with the expression of 3-hydroxy-3-methylglutaryl-CoAreductase (HMRCG), and citrullinated histone H3 (H3cit), in tissues from breast cancer patients. High expression of CCDC25 and HMGCR was related with worse prognosis in breast cancer patients. In conclusion, our study explores a novel mechanism for de novo cholesterol biosynthesis in the regulation of CCDC25 expression, NETs formation and breast cancer metastasis. Targeting cholesterol biosynthesis may be promising therapeutic strategies to treat breast cancer metastasis.Magnetic Resonance Imaging (MRI) provides a unique opportunity to investigate neural changes in healthy and clinical conditions. Its large inherent susceptibility to motion, however, often confounds the measurement. Approaches assessing, correcting, or preventing motion corruption of MRI measurements are under active development, and such efforts can greatly benefit from carefully controlled datasets. We present a unique dataset of structural brain MRI images collected from 148 healthy adults which includes both motion-free and motion-affected data acquired from the same participants. This matched dataset allows direct evaluation of motion artefacts, their impact on derived data, and testing approaches to correct for them. Our dataset further stands out by containing images with different levels of motion artefacts from the same participants, is enriched with expert scoring characterizing the image quality from a clinical point of view and is also complemented with standard image quality metrics obtained from MRIQC. The goal of the dataset is to raise awareness of the issue and provide a useful resource to assess and improve current motion correction approaches.Extraordinary heat extremes occurred in the 1930s in areas of the Northern Hemisphere far from the record setting heat over the US associated with the Great Plains Dust Bowl drought. A climate model sensitivity experiment is used to identify a new mechanism involving a warm season circumglobal atmospheric teleconnection pattern that spread heat extremes over far-flung areas of the Northern Hemisphere arising from the intense heating over the desiccated Great Plains themselves. It has only been in the twenty-first century that human populations in these regions of the Northern Hemisphere have experienced heat extremes comparable to the 1930s. This demonstrates that humans influenced Northern Hemisphere temperature and heat extremes through disastrous and unprecedented regional land use practices over the Great Plains, and points to the possibility that future intense regional droughts could affect heat extremes on hemispheric scales.To investigate the delayed release characteristics of CO gas in the initial stage of the low-temperature oxidation of coal, closed oxygen consumption experiments were conducted on coal samples taken from the Hongqingliang coal mine, and the corresponding relationship between the CO concentration and time in the initial stage of the experimental reaction was analyzed. A physical adsorption model of the macromolecules in coal for O2 and CO was established, and the difference in the competitive adsorption between the CO and O2 gas molecules on the coal surface was analyzed from a microscopic perspective using the grand canonical ensemble Monte Carlo simulation. The results showed a delayed CO release phenomenon in the initial stage of the reaction in all the experiments, and the delayed time of CO release was negatively correlated with the temperature; the relationship between the adsorption amounts of CO and O2 in the molecular structure model of coal was CO > O2. With increasing temperature, the adsorption capacity of the two gases decreased. Under the same conditions, there was competitive adsorption of the mixture of CO and O2 by coal, with the adsorption capacity of CO being much greater than that of O2. The adsorption of CO gas molecules by coal played an inhibitory role in the release of CO gas in the initial oxidation stage. The study results are expected to help understand the CO generation characteristics in the goaf of coal seam working faces and thus prevent coal mine disasters.Comparisons are made between six different approved face masks concerning their particle transmissibility allied to mechanical properties. The latter involves material testing and stretch or strain behaviour under load. SEM and X-ray elemental analyses showed contrasting structures between random and ordered fibre orientations. These constitute the mask designs where transmissibility is to be minimised. Airflow velocity measurement enabled filtration to be measured between the different mask designs, from two to six layers of different fabrics in combination. SEM provided the fibre diameter and pore size of each mask layer, up to a maximum of six. Stretching each complete mask showed its elasticity and recovery behaviour on an energy basis. The energy conversion involved in mask straining involves areas enclosed within steady and cyclic load-extension plots. Thus, the work done in extending a mask and the energy recovered from its release identified a hysteresis associated with an irrecoverable permanent stretch to the mask fabric. Failure of individual layers, which occurred successively in extended stretch tests, appeared as a drop in a load-extension response. That change is associated with permanent damage to each mask and friction contact within the rearrangement of loose fibre weaves. Masks with the greatest number of layers reduced particle transmissibility. However, woven or ordered mask fabrics in two layers with different orientations provided comparable performance. Simulation of each mechanical response, velocity streamlining and fibre distribution within the mask layers are also presented.
