Recent genetic research has explored how genetic variants may contribute to gender dysphoria and transgender and gender-diverse (TGD) identities. When investigating communities that have been marginalized, it is important for researchers to incorporate perspectives of the communities the research is targeting. Therefore, investigators should incorporate the TGD community's opinions into this research to mitigate potential ethical issues, given the history of pathologization of TGD identities and utilization of genetics for eugenics. The aim of this study was to understand the perspectives of TGD individuals about trans-associated genetic research (TAGR). Eighteen semi-structured interviews were conducted with members of the TGD community to explore how TGD individuals view TAGR. Through inductive content analysis, five major themes were emergent (1) TAGR could affect self-perception of identity; (2) TAGR could affect external views of TGD people; (3) TAGR could affect access to gender-affirming services; (4) TAGR could contribute to the pathologization and elimination of TGD identities; and (5) researchers should consult TGD community members and consider ethical concerns before conducting research. Participants highlighted concerns about TAGR being used as a tool for discrimination. With rapidly evolving computational tools and methods, QSP modeling has achieved important progress in pharmaceutical research and development, including for heart failure (HF). link2 However, various challenges exist in the QSP modeling and clinical characterization of HF. Machine/deep learning (ML/DL) methods have had success in a wide variety of fields and disciplines. They provide data-driven approaches in HF diagnosis and modeling, and offer a novel strategy to inform QSP model development and calibration. The combination of ML/DL and QSP modeling becomes an emergent direction in the understanding of HF and clinical development new therapies. In this work, we review the current status and achievement in QSP and ML/DL for HF, and discuss remaining challenges and future perspectives in the field.Blocking the main replicating enzyme, 3 Chymotrypsin-like protease (3CLpro) is the most promising drug development strategy against the SARS-CoV-2 virus, responsible for the current COVID-19 pandemic. In the present work, 9101 drugs obtained from the drug bank database were screened against SARS-CoV-2 3CLpro prosing deep learning, molecular docking, and molecular dynamics simulation techniques. In the initial stage, 500 drug-screened by deep learning regression model and subjected to molecular docking that resulted in 10 screened compounds with strong binding affinity. Further, five compounds were checked for their binding potential by analyzing molecular dynamics simulation for 100 ns at 300 K. In the final stage, two compounds 4-[(2s,4e)-2-(1,3-Benzothiazol-2-Yl)-2-(1h-1,2,3-Benzotriazol-1-Yl)-5-Phenylpent-4-Enyl]Phenyl(Difluoro)Methylphosphonic Acid and 1-(3-(2,4-dimethylthiazol-5-yl)-4-oxo-2,4-dihydroindeno[1,2-c]pyrazol-5-yl)-3-(4-methylpiperazin-1-yl)urea were screened as potential hits by analyzing several parameters like RMSD, Rg, RMSF, MMPBSA, and SASA. Thus, our study suggests two potential drugs that can be tested in the experimental conditions to evaluate the efficacy against SARS-CoV-2. Further, such drugs could be modified to develop more potent drugs against COVID-19.
The higher prevalence of cognitive impairment/ dementia among cancer survivors is likely multifactorial. Since both exposures to cytomegalovirus (CMV) and inflammation are common among elderly cancer survivors, we evaluated their contribution towards dementia.
Data from 1387 cancer survivors and 7004 participants without cancer in the 2016 wave of the Health and Retirement Study (HRS) was used in this study. Two inflammatory biomarkers, C-reactive protein (CRP) and neutrophil-lymphocyte ratio (NLR), were used to create an inflammation score. We used survey logistic regression adjusted for survey design parameters.
CMV seropositivity was not associated with cognitive impairment among cancer survivors (p = 0.2). In addition, inflammation was associated with elevated odds of cognitive impairment (OR = 2.2, 95% CI [1.2, 4.2]). Cancer survivors who were both CMV seropositive and had increased inflammation had the highest odds of cognitive impairment compared to those who were CMV seronegative and had low inftreatment may be important in reducing the prevalence of cognitive impairment/ dementia among cancer survivors.
Folate and vitamin B12 deficiencies can impair proper growth and brain development in children. Data on the folate and vitamin B12 status of children aged 6-59months in Guatemala are scarce. Identification of factors associated with higher prevalence of these micronutrient deficiencies within the population is needed for national and regional policymakers.
To describe national and regional post-fortification folate and vitamin B12 status of children aged 6-59months in Guatemala.
A multistage, cluster probability study was carried out with national and regional representation of children aged 6-59months. Demographic and health information was collected for 1246 preschool children, but blood samples for red blood cell (RBC) folate and vitamin B12 were collected and analyzed for 1,245 and 1143 preschool children, respectively. We used the following deficiency criteria as cutoff points for the analyses < 305nmol/L for RBC folate, < 148pmol/L for vitamin B12 deficiency, and 148-221pmol/L for marginal voorest families. These findings suggest gaps in the coverage of fortification and the need for additional implementation strategies to address these gaps in coverage to help safeguard the health of Guatemalan children.
In this study, a third of all children had RBC folate deficiency and half were vitamin B12 deficient. Persistent disparities in breastfeeding rates among African American (AA) women compared to other population groups have motivated researchers to understand factors influencing breastfeeding choices using a variety of methods. Quantitative surveys are more commonly reported, however, qualitative work that amplifies voices of AA women is limited.
Participants were recruited from a randomized controlled feasibility trial focused on breastfeeding support for AA women in Detroit, MI. Thirteen women were enrolled in the qualitative portion of the study described here. Using the Socioecological model (SEM) as the theoretical foundation, semi-structured qualitative interviews were conducted to explore perceived facilitators and barriers to breastfeeding. Interviews were digitally recorded, transcribed, and analyzed using Theoretical thematic analysis.
Women reported factors ranging from micro to macro SEM levels that discouraged or reinforced breastfeeding. Key challenges included breastfeeding-related discouraborative discussions between women and healthcare providers focused on suggestions provided by AA women should be encouraged.
State medicaid programs provide access to effective contraception for people with lower incomes. This study examined contraception use and pregnancy among reproductive-age women enrolled in the South Carolina Medicaid, by eligibility program and socio-demographic sub-groups.
A retrospective cohort of women aged 15-45 who were newly eligible for South Carolina Medicaid from 2012 to 2016 was examined. Log-binomial regression and average marginal effects assessed relationships between contraception use and pregnancies ending in live and non-live births. Contraception was categorized as permanent, long acting reversible contraception (LARC), short-acting hormonal contraception (SAC), or no contraceptive claims. Women with family planning or full-benefit medicaid coverage were included.
Approximately 11% of women used LARC methods, 41% used SAC methods, and 46% had no evidence of contraceptive claims. Method utilization varied by eligibility program, race/ethnicity and age. The likelihood of pregnancy was loof pregnancy was lower among women using SAC methods and markedly lower among women using LARC. link3 Variation in contraceptive use among racial/ethnic groups was noted despite Medicaid coverage. As new policies and initiatives emerge, these findings provide important context for understanding the role of Medicaid programs in reducing financial barriers to contraceptive services and ensuring access to effective contraception, while fostering reproductive health autonomy among women.
Vitreoretinal surgery in eyes with Boston type 1 keratoprosthesis (KPro) is challenging due to narrow optic of the KPro. This study analyzed the results of pars plana vitrectomy (PPV) using a wide-field imaging accessory, Resight®700 Fundus Viewing System (Carl Zeiss Meditec, Inc., Germany), for better intraoperative peripheral retinal imaging.
In this retrospective case series, KPro patients who underwent simultaneous or sequential PPV at Dokuz Eylul University Hospital between June 2010 and January 2020 were evaluated in terms of anatomic and visual prognoses, as well as KPro- and PPV-associated complications.
Among 9 KPro eyes that necessitated vitreoretinal surgery, 3 (33.3%) underwent simultaneous KPro and PPV due to proliferative vitreoretinopaties; 6 (66.7%) underwent PPV for retinal detachment or suprachoroidal hemorrhage that appeared after KPro surgery. Retina could be attached in 7 eyes (77.8%), and vision improved in 3 eyes (33.3%). In 1 eye, injected silicone oil moved to subconjunctival area through glaucoma drainage device.
In eyes with a Boston KPro, wide-angle viewing systems helped handling peripheral retinal problems successfully during PPV, with no observed inadequacy of imaging. Despite anatomical success in most cases, visual prognosis depends on vitality of the macula and the optic disc.
