Complete structural assignment of glucosylated products revealed that the bioconversion by regio-selective glucosylation of cortisone and prednisone produced cortisone 21-glucoside and prednisone 21-glucoside, respectively. We also combined molecular dynamics (MD) simulation to study the binding feature and mechanism of glucosylation. MD simulation studies showed the formation of a stable complex between protein, glucose donor, and substrate, stabilized by hydrogen bonds. Fluorescence spectra analysis showed the fluorescence of Hb was statically quenched by HANs in the sequence of TCAN > DBAN > DCAN > IAN > BAN > CAN. HANs could combine to multiple surface sites of Hb accounting for "hydrogen bonds" and "van der Waals forces". The high-resolution mass spectra analysis for Hb with and without HANs further confirmed the formation of multiple HAN-Hb complexes with different conversion rates. With the assistance of MOE molecule docking, the potential combination sites and related interactions parameters between HANs and Hb were filtrated. By analyzing the correlations between the candidate interactions parameters and fluorescence quenching constants/MS conversion rates, the combination sites of HANs were fixed at Asp126 (α1/α2), Lys127 (α1/α2) in the form of "hydrogen bonds" X → Asp126 (α1/α2), N → Lys127 (α1/α2). In this way, the potential interactions between HANs and Hb were effectively evaluated.
Attention Deficit Hyperactivity Disorder (ADHD) is a neurodevelopmental disorder that affects approximately 2-7 % of children globally and is associated with a myriad of difficulties that have long-term consequences. Most children and adolescents live in low- and middle-income countries (LMICs), but there are few reports and no consolidation of findings on ADHD treatment outcomes in this population. We conducted a review of ADHD treatment literature for children and adolescents living in LMICs.
Studies were identified using databases (PsychoINFO, Pubmed, MEDLINER, EMBASE, Global Health, Academic Search Complete, Google Scholar). The initial search produced 139 articles. These were filtered for language, title, abstract, and full-text keyword identification to yield a final 20 articles to be included in this review.

It causes life-threatening infections, such as ventilator-associated pneumonia (VAP), bacteremia, meningitis, wound and urinary tract infections (UTI). Increase in carbapenem resistance exhibited by A. baumannii has accentuated the need for novel targets for effective treatment. Despite the pronounced relevance of PPK2 as a pathogenicity determinant in several pathogens, it has not been explored as a drug target in A. baumannii. The present study was piloted to investigate the substrate binding by A. baumannii PPK2 (AbPPK2), a two-domain Class II polyphosphate kinase 2. A homology model of AbPPK2 was developed and validated for molecular docking of ATP and ADP in the predicted binding pocket. Further analysis of AbPPK2 revealed a set of common residues in the catalytic cleft interacting with ATP and ADP which would be useful for the screening of inhibitors against A. baumannii.
Ultra long-term monitoring with subcutaneous EEG (sqEEG) offers objective outpatient recording of electrographic seizures as an alternative to self-reported epileptic seizure diaries. This methodology requires an algorithm-based automatic seizure detection to indicate periods of potential seizure activity to reduce the time spent on visual review. The objective of this study was to evaluate the performance of a sqEEG-based automatic seizure detection algorithm.
A multicenter cohort of subjects using sqEEG were analyzed, including nine people with epilepsy (PWE) and 12 healthy subjects, recording a total of 965days. The automatic seizure detections of a deep-neural-network algorithm were compared to annotations from three human experts.
Data reduction ratios were 99.6% in PWE and 99.9% in the control group. The cross-PWE sensitivity was 86% (median 80%, range 69-100% when PWE were evaluated individually), and the corresponding median false detection rate was 2.4 detections per 24 hours (range 2.0-13.0).
Our findings demonstrated that step one in a sqEEG-based semi-automatic seizure detection/review process can be performed with high sensitivity and clinically applicable specificity.

Comprehensive Behavioral Intervention for Tics (CBIT) is a first-line treatment of Tourette syndrome (TS). However, the brain mechanisms involved in CBIT are poorly understood. Enhanced frontomesial EEG coherence during a Go/NoGo task has been suggested as a mechanism involved in voluntary tic control. In the current study, we conducted a randomized controlled trial to assess whether EEG coherence during a Go/NoGo task was associated with CBIT outcome.
Thirty-two children with TS were randomly assigned to CBIT or to treatment-as-usual (TAU). Treatment outcome was assessed by a blinded evaluator with the Yale Global Tic Severity Scale (YGTSS) and the Clinical Global Impression - Improvement Scale (CGI-I). EEG was recorded during a Go/NoGo task at baseline and endpoint. EEG coherence was computed in the alpha frequency band between a priori selected channel pairs spanning the frontal and motor areas.
Tic severity decreased significantly in the CBIT group. However, CBIT did not impact EEG coherence and baseline EEG coherence did not predict treatment outcome.
Although CBIT was superior to TAU on blinded clinical outcomes, EEG coherence during the Go/NoGo task was not associated with change in tic severity.

The phase of gonad maturity was based on histological analysis and the gonadosomatic index. All fish had properly developed gonads in the pre-spawning, spawning and late spawning phases of the reproductive cycle in the fish collected in April, June and October, respectively. Diploid and triploid females and males did not differ in mean GSI in all reproductive phases. In October, ovaries of most females contained vitellogenic oocytes, testes had large numbers of spermatozoa. The hormone concentrations in both tissues depended mainly on the reproductive phase, but not on ploidy. The patterns of changes in E2 concentrations was similar in females and males. In the pre-spawning phase, diploid and triploid females showed differences in the concentrations of 11-KT in gonads and plasma, and T in plasma. Diploid males differed in pattern of T concentrations in gonads and plasma, while all males showed a similar pattern of 11-KT plasma concentrations. Spermatozoa observed in triploid males suggest that they participate in reproduction. Gibel carps, regardless of ploidy, had an extended period of reproduction, which makes this invasive species a potentially greater threat to native ichthyofauna. Here we report on the largest study to date of the accuracy, reproducibility (inter-examiner variation), and repeatability (intra-examiner variation) of footwear examiners' decisions. In this study, 84 practicing footwear examiners each conducted up to 100 comparisons between questioned footwear impressions (provided as photographs and digital images) and known footwear (provided as photographs, transparent test impressions, and digital images), resulting in a total of 6610 comparisons. The quality and characteristics of the impressions were selected to be broadly representative of those encountered in casework. Glioblastoma (GBM) is the most common malignancy tumor of central nervous system. PTBP3 was closely associated with the development of tumor. However, the function and molecular mechanism of PTBP3 in GBM is little known.
qPCR and immunoblotting were used to detect PTBP3 expression levels in glioma tissues and cells. CCK8, Edu, flow cytometry, wound healing, and transwell assays were used to examined the function of PTBP3 in GBM. qPCR, Immunoblotting, and ubiquitination assays were performed to identify the mechanism of PTBP3.
We found that PTBP3 was upregulated in GBM, and high expression of PTBP3 correlated with the poor survival of GBM patients. PTBP3 knockdown reduced proliferation, invasion, and migration of GBM. Conversely, overexpressing PTBP3 has an opposite effect. Moreover, PTBP3 had an effect on the EMT of GBM. More importantly, we found that PTBP3 stabilized Twist1 by decreasing its ubiquitination and degradation. Furthermore, orthotopic xenograft models were used to demonstrate the PTBP3 on the development of GBM in vivo.
