In this review, we propose to analyze in particular the new developments in multiplexed approaches to miRNA detection. The main aspects of these methods (including sensitivity and specificity) will be analyzed, with a particular focus on the demonstrated multiplexing capability and potential of each of these methods.Carbohydrate response element-binding protein (ChREBP) is critical in the regulation of fatty acid and triglyceride synthesis in the liver. Interestingly, Chrebp-/- mice show reduced levels of plasma cholesterol, which is critical for steroid hormone synthesis in adrenal glands. Furthermore, Chrebp mRNA expression was previously reported in human adrenal glands. Thus, it remains to be investigated whether ChREBP plays a role directly or indirectly in steroid hormone synthesis and release in adrenal glands. In the present study, we find that Chrebp mRNA is expressed in mouse adrenal glands and that ChREBP binds to carbohydrate response elements. Histological analysis of Chrebp-/- mice shows no adrenal hyperplasia and less oil red O staining compared with that in WT mice. In adrenal glands of Chrebp-/- mice, expression of Fasn and Scd1, two enzymes critical for fatty acid synthesis, was substantially lower and triglyceride content was reduced. Expression of Srebf2, a key transcription factor controlling synthesis and uptake of cholesterol and the target genes, was upregulated, while cholesterol content was not significantly altered in the adrenal glands of Chrebp-/- mice. Adrenal corticosterone content and plasma adrenocorticotropic hormone and corticosterone levels were not significantly altered in Chrebp-/- mice. Consistently, expression of genes related to steroid hormone synthesis was not altered. Corticosterone secretion in response to two different stimuli, namely 24-h starvation and cosyntropin administration, was also not altered in Chrebp-/- mice. Taking these results together, corticosterone synthesis and release were not affected in Chrebp-/- mice despite reduced plasma cholesterol levels.
Mobile health (mHealth) apps play an important role in delivering education, providing advice on treatment, and monitoring patients' health. Good usability of mHealth apps is essential to achieve the objectives of mHealth apps efficiently. To date, there are questionnaires available to assess the general system usability but not explicitly tailored to precisely assess the usability of mHealth apps. Hence, the mHealth App Usability Questionnaire (MAUQ) was developed with 4 versions according to the type of app (interactive or standalone) and according to the target user (patient or provider). Standalone MAUQ for patients comprises 3 subscales, which are ease of use, interface and satisfaction, and usefulness.

This study aimed to translate and validate the English version of MAUQ (standalone for patients) into a Malay version of MAUQ (M-MAUQ) for mHealth app research and usage in future in Malaysia.

Forward and backward translation and harmonization of M-MAUQ were conducted by Malay native speakers who alin the future.
Urinary incontinence (UI) and lower urinary tract symptoms (LUTS) are commonly experienced by adult patients in hospitals (inpatients). Although peak bodies recommend that health services have systems for optimal UI and LUTS care, they are often not delivered. For example, results from the 2017 Australian National Stroke Audit Acute Services indicated that of the one-third of acute stroke inpatients with UI, only 18% received a management plan. In the 2018 Australian National Stroke Audit Rehabilitation Services, half of the 41% of patients with UI received a management plan. There is little reporting of effective inpatient interventions to systematically deliver optimal UI/LUTS care.

This study aims to determine whether our UI/LUTS practice-change package is feasible and effective for delivering optimal UI/LUTS care in an inpatient setting. The package includes our intervention that has been synthesized from the best-available evidence on UI/LUTS care and a theoretically informed implementation strategy ation and are undertaking postimplementation data collection (T

Our practice-change package is designed to reduce the current inpatient UI/LUTS evidence-based practice gap, such as those identified through national stroke audits. Due to the rapid digitalization of health care, leadership is becoming more complex. Leadership in digital health services is a term that has been used in the literature with various meanings. Conceptualization of leadership in digital health services is needed to deliver higher quality digital health services, update existing leadership practices, and advance research.

The aim of this study is to outline a concept analysis that aims to clarify and define the concept of leadership in digital health services.

Health-enabling technologies (HETs) are information and communication technologies that promote individual health and well-being. An important application of HETs is telerehabilitation for patients with musculoskeletal shoulder disorders. Currently, there is no overview of HETs that assist patients with musculoskeletal shoulder disorders when exercising at home.

This scoping review provides a broad overview of HETs that assist patients with musculoskeletal shoulder disorders when exercising at home. It focuses on concepts and components of HETs, exercise program strategies, development phases, and reported outcomes.

The search strategy used Medical Subject Headings and text words related to the terms upper extremity, exercises, and information and communication technologies. The MEDLINE, Embase, IEEE Xplore, CINAHL, PEDro, and Scopus databases were searched. Two reviewers independently screened titles and abstracts and then full texts against predefined inclusion and exclusion criteria. A systematic narrative synthesis was performed.

NH, resident, and PrI characteristics across race/ethnicity groups were compared using analysis of variance and chi-square. Participants were 62% female and 89% functionally dependent. More Black and Asian individuals had peripheral vascular disease. More Black individuals had persistent trunk and Stage 4 PrIs. Black and Hispanic individuals had normal skin color surrounding PrIs. More Asian individuals had PrIs surrounded by purple/red discolored skin. More Black individuals' heel PrIs were unstageable, necrotic, and showed no granulation. The four domains, derived from factor analysis, affirmed the supportive perception among the public (score range = 4.42 to 4.64/5.0). However, among the gaps identified were the perception toward hand sanitizing and susceptibility of older adults to COVID-19. Public knowledge on symptoms of COVID-19 was limited (e.g., 41.4% to 53.4% incorrect responses to anosmia, diarrhea, confusion). Multivariate analysis of variance found that males and participants with less education had significantly lower supportive perceptions (p 86% of participants having access to internet communication technology, mobile interventions tailored to gender and educational level are recommended to promote long-term pandemic preparedness among stakeholders and the public. Data are reported for five adults with dementia receiving day services at an all-inclusive care center for older adults (one male; mean age = 82.8 years). Data were collected three times to obtain baseline (Time 1 to Time 2) and post-intervention (Time 2 to Time 3) scores. Self-reported quality of life and depression symptoms improved post-intervention, but not during baseline. Caregiver-reported agitation did not change consistently across participants during baseline or post-intervention. Simple ownership of affordable robotic pets may be a feasible option to improve quality of life and affective experiences in individuals with dementia. Further work with larger samples should examine whether a combination of ownership and structured activity would produce changes more perceptible to caregivers. Colletotrichum gloeosporioides is one of the most common and serious fungal diseases of the tree Mangifera persiciforma. Yet we lack an effective method to evaluate this ecological interaction accurately. Here, we measured the functional traits and leaf reflectance spectrum of the host plants under different disease degrees. The findings provide a fast and efficient method for large-scale and high-precision monitoring of C. gloeosporioides in M. persiciforma stands. Using the collected leaf reflection data, we set up a prediction model of the optimal disease degree. Firstly, we found that leaf functional traits of M. persiciforma generally consisted of low leaf thickness, low relative chlorophyll content, small specific leaf area, high leaf tissue density, high dry matter content, low stomatal density, and large stomatal area. Secondly, leaf reflectivity increases with damage of C. gloeosporioides, which corresponds to five main reflection peaks and five absorption valleys in the spectral reflectance curve of leaves at the same positions (350-1800 nm). Thirdly, with the increase of infection degree, red edge slope and yellow edge slope decrease, while green peak reflectance, red valley reflectance, and blue edge slope all increase. Blue shift was detected in the red edge, green peak, and red valley, while red shift appeared at the blue edge and yellow edge. Finally, the best predictive model was that based on green peak reflectance (y=3.6396-0.0693x, R2=0.5149, RMSE [root-mean-square error] =0.2735), with an R2=0.92 and RMSE=0.0042 between its predicted vs. observed values. Because of its high inversion accuracy, the model can be used to predict the invasion conditions of M. persiciforma by C. gloeosporioides. Our study demonstrated that when plants are infected by C. gloeosporioides, there was a strong trade-off relationship between leaf functional traits. On the global leaf economics spectrum, the leaves tended toward the "slow investment-return" end when infected by C. gloeosporioides.Deforestation and forest degradation are among the leading global concerns, as they could reduce the carbon sink and sequestration potential of the forest. The impoundment of Kenyir River, Hulu Terengganu, Malaysia, in 1985 due to the development of hydropower station has created a large area of water bodies following clearance of forested land. This study assessed the loss of forest carbon due to these activities within the period of 37 years, between 1972 and 2019. The study area consisted of Kenyir Lake catchment area, which consisted mainly of forests and the great Kenyir Lake. Remote sensing datasets have been used in this analysis. Satellite images from Landsat 1-5 MSS and Landsat 8 OLI/TRIS that were acquired between the years 1972 and 2019 were used to classify land uses in the entire landscape of Kenyir Lake catchment. https://www.selleckchem.com/products/molidustat-(bay85-3934).html Support vector machine (SVM) was adapted to generate the land-use classification map in the study area. The results show that the total study area includes 278,179 ha and forest covers dominated the area for before and after the impoundment of Kenyir Lake. The main single cause attributing to the forest loss was due to clearing of forest for hydro-electric dam construction. However, the remaining forests surrounding the study area are still able to sequester carbon at a considerable rate and thus balance the carbon dynamics within the landscapes. The results highlight that carbon sequestration scenario in Kenyir Lake catchment area shows the potential of the carbon sink in the study area are acceptable with only 17% reduction of sequestration ability. The landscape of the study area is considered as highly vegetated area despite changes due to dam construction.Monosodium glutamate (MSG) is a common flavor enhancer and stabilizer for ready-made or packaged foods. This research investigated the impact of MSG on the maternal and fetal liver. The present study was carried out on sixteen mature female Albino rats and eight male rats of reproductive age. The control group was dissected on day 20 of gestation. MSG group was administrated MSG daily at a dosage of 1 g/5 mL/kg body weight from day 0 to day 20 of gestation. The liver function and lipid profile of the control and treated mothers were investigated in the blood sera. The levels of nitric oxide (NO), tumor necrosis factor (TNF-α), superoxide dismutase (SOD), and reduced glutathione (GSH) activities in the liver homogenate of maternal and fetal tissue were assayed, in addition to histopathological, histochemical and immunohistochemical studies were done to the liver tissue. The activities of liver functions and lipid profile significantly altered in the treated mothers with MSG. MSG significantly reduced the SOD and reduced GSH activities in addition to the elevated TNF-α and NO in liver tissue of pregnant mothers and their fetuses. Severe histopathological alterations were observed in both maternal and fetal liver tissues of MSG-treated groups. Moreover, histochemical observations showed a reduction of total polysaccharides in the liver of pregnant rats and fetuses. A significant increase in the percentage area of positive immunoreaction for caspase 3 was observed in the liver of treated rats with MSG compared to the liver of the control. The liver of fetuses treated with MSG revealed an alteration like their mother. This study showed that during the gestational period MSG exposure resulted in several biochemical, histological, and histochemical changes in the maternal and fetal liver tissues which emphasize the toxic effect of MSG.In this paper, three bioretention facilities (BT, RG1-A, and RG1-B) were selected for on-site testing and experimental analysis. Of which, BT is a roadside bioretention tank with layered filler, while RG1-A and RG1-B are rain gardens with conventional filler (Bioretention soil media, BSM) and modified filler (BSM+10% Water treatment residuals,WTR), respectively. The effect of pollutant accumulation on the soil microbial community structure in the facilities, and the risk of heavy metal contamination over several years of bioretention facility operation were studied. Results showed that the water quality pollutant load reduction in BT was fluctuating. This is related to the poor water quality of road stormwater flowing into BT and the facility filler. Because RG1-B uses modified filler, RG1-B was more effective than RG1-A in regulating water quality and quantity; the changes in soil physical and chemical properties in BT, RG1-A, and RG1-B were influenced by external factors. Next, BT was at high risk of heavy metal contamination than other facilities.

The emergence of neuropsychiatric symptoms may be one indicator of early AD-related cognitive decline, but distinguishing symptoms from those due to other causes can be challenging. Mild behavioral impairment (MBI) describes an at-risk state for cognitive decline characterized by the late-life onset of neuropsychiatric symptoms. In this article, we discuss the current conceptualization of MBI, the potential for its characteristic neuropsychiatric symptoms to indicate risk for future cognitive decline, and present potential clinical implications. [Journal of Gerontological Nursing, 47(3), 29-36.].The current study examines the public's perception on the mitigation of COVID-19 risk and knowledge of disease symptoms among older adults in nursing homes, with the intention to address gaps in knowledge using mobile technologies. An online survey questionnaire was completed by 611 adults residing in Malaysia. The four domains, derived from factor analysis, affirmed the supportive perception among the public (score range = 4.42 to 4.64/5.0). However, among the gaps identified were the perception toward hand sanitizing and susceptibility of older adults to COVID-19. Public knowledge on symptoms of COVID-19 was limited (e.g., 41.4% to 53.4% incorrect responses to anosmia, diarrhea, confusion). Multivariate analysis of variance found that males and participants with less education had significantly lower supportive perceptions (p 86% of participants having access to internet communication technology, mobile interventions tailored to gender and educational level are recommended to promote long-term pandemic preparedness among stakeholders and the public. Data are reported for five adults with dementia receiving day services at an all-inclusive care center for older adults (one male; mean age = 82.8 years). Data were collected three times to obtain baseline (Time 1 to Time 2) and post-intervention (Time 2 to Time 3) scores. Self-reported quality of life and depression symptoms improved post-intervention, but not during baseline. Caregiver-reported agitation did not change consistently across participants during baseline or post-intervention. Simple ownership of affordable robotic pets may be a feasible option to improve quality of life and affective experiences in individuals with dementia. Further work with larger samples should examine whether a combination of ownership and structured activity would produce changes more perceptible to caregivers. Colletotrichum gloeosporioides is one of the most common and serious fungal diseases of the tree Mangifera persiciforma. Yet we lack an effective method to evaluate this ecological interaction accurately. Here, we measured the functional traits and leaf reflectance spectrum of the host plants under different disease degrees. The findings provide a fast and efficient method for large-scale and high-precision monitoring of C. gloeosporioides in M. persiciforma stands. Using the collected leaf reflection data, we set up a prediction model of the optimal disease degree. Firstly, we found that leaf functional traits of M. persiciforma generally consisted of low leaf thickness, low relative chlorophyll content, small specific leaf area, high leaf tissue density, high dry matter content, low stomatal density, and large stomatal area. Secondly, leaf reflectivity increases with damage of C. gloeosporioides, which corresponds to five main reflection peaks and five absorption valleys in the spectral reflectance curve of leaves at the same positions (350-1800 nm). Thirdly, with the increase of infection degree, red edge slope and yellow edge slope decrease, while green peak reflectance, red valley reflectance, and blue edge slope all increase. Blue shift was detected in the red edge, green peak, and red valley, while red shift appeared at the blue edge and yellow edge. Finally, the best predictive model was that based on green peak reflectance (y=3.6396-0.0693x, R2=0.5149, RMSE [root-mean-square error] =0.2735), with an R2=0.92 and RMSE=0.0042 between its predicted vs. observed values. Because of its high inversion accuracy, the model can be used to predict the invasion conditions of M. persiciforma by C. gloeosporioides. Our study demonstrated that when plants are infected by C. gloeosporioides, there was a strong trade-off relationship between leaf functional traits. On the global leaf economics spectrum, the leaves tended toward the "slow investment-return" end when infected by C. gloeosporioides.Deforestation and forest degradation are among the leading global concerns, as they could reduce the carbon sink and sequestration potential of the forest. The impoundment of Kenyir River, Hulu Terengganu, Malaysia, in 1985 due to the development of hydropower station has created a large area of water bodies following clearance of forested land. This study assessed the loss of forest carbon due to these activities within the period of 37 years, between 1972 and 2019. The study area consisted of Kenyir Lake catchment area, which consisted mainly of forests and the great Kenyir Lake. Remote sensing datasets have been used in this analysis. The results show that the total study area includes 278,179 ha and forest covers dominated the area for before and after the impoundment of Kenyir Lake. The main single cause attributing to the forest loss was due to clearing of forest for hydro-electric dam construction. However, the remaining forests surrounding the study area are still able to sequester carbon at a considerable rate and thus balance the carbon dynamics within the landscapes. The results highlight that carbon sequestration scenario in Kenyir Lake catchment area shows the potential of the carbon sink in the study area are acceptable with only 17% reduction of sequestration ability. This research investigated the impact of MSG on the maternal and fetal liver. The present study was carried out on sixteen mature female Albino rats and eight male rats of reproductive age. The control group was dissected on day 20 of gestation. MSG group was administrated MSG daily at a dosage of 1 g/5 mL/kg body weight from day 0 to day 20 of gestation. The liver function and lipid profile of the control and treated mothers were investigated in the blood sera. The levels of nitric oxide (NO), tumor necrosis factor (TNF-α), superoxide dismutase (SOD), and reduced glutathione (GSH) activities in the liver homogenate of maternal and fetal tissue were assayed, in addition to histopathological, histochemical and immunohistochemical studies were done to the liver tissue. The activities of liver functions and lipid profile significantly altered in the treated mothers with MSG. MSG significantly reduced the SOD and reduced GSH activities in addition to the elevated TNF-α and NO in liver tissue of pregnant mothers and their fetuses. Severe histopathological alterations were observed in both maternal and fetal liver tissues of MSG-treated groups. Moreover, histochemical observations showed a reduction of total polysaccharides in the liver of pregnant rats and fetuses. A significant increase in the percentage area of positive immunoreaction for caspase 3 was observed in the liver of treated rats with MSG compared to the liver of the control. The liver of fetuses treated with MSG revealed an alteration like their mother. This study showed that during the gestational period MSG exposure resulted in several biochemical, histological, and histochemical changes in the maternal and fetal liver tissues which emphasize the toxic effect of MSG.In this paper, three bioretention facilities (BT, RG1-A, and RG1-B) were selected for on-site testing and experimental analysis. Of which, BT is a roadside bioretention tank with layered filler, while RG1-A and RG1-B are rain gardens with conventional filler (Bioretention soil media, BSM) and modified filler (BSM+10% Water treatment residuals,WTR), respectively. The effect of pollutant accumulation on the soil microbial community structure in the facilities, and the risk of heavy metal contamination over several years of bioretention facility operation were studied. Results showed that the water quality pollutant load reduction in BT was fluctuating. This is related to the poor water quality of road stormwater flowing into BT and the facility filler. Because RG1-B uses modified filler, RG1-B was more effective than RG1-A in regulating water quality and quantity; the changes in soil physical and chemical properties in BT, RG1-A, and RG1-B were influenced by external factors. Next, BT was at high risk of heavy metal contamination than other facilities.

Our results show that low concentration of WV has protective effects on the oxidative damage caused by PFOS in the planarian.The developing world is facing pivotal challenges in recent times. Among these, global warming has ominous repercussions on every segment of society, thus tracing its underlying causes is imperative. This research attempts to investigate the impact of urbanization and energy consumption on carbon dioxide emissions (CO2) for a panel of 8 Asian countries (Bangladesh, China, India, Indonesia, Malaysia, Nepal, Pakistan, and Sri Lanka) over the period 1982 to 2017. The analyses are executed using panel co-integration and Granger causality techniques. The main findings of panel co-integration reveal a long-run relationship between urbanization, energy consumption, and CO2 emissions. Furthermore, the results indicate a positive and significant impact of urbanization and energy consumption on CO2 emissions, indicating that urban development and high energy consumptions are barriers to improve environmental quality in the long run. The results also highlight bi-directional causality between energy consumption and urbanization, while unidirectional causality exists between energy consumption and CO2 emissions. Based on the obtained results, this study offers useful policy implications for plummeting carbon emissions.Ambitious energy targets in the 2020 European climate and energy package have encouraged many stakeholders to explore and implement measures improving the energy efficiency of water and wastewater treatment facilities. Model-based process optimization can improve the energy efficiency of wastewater treatment plants (WWTP) with modest investment and a short payback period. However, such methods are not widely practiced due to the labor-intensive workload required for monitoring and data collection processes. This study offers a multi-step simulation-based methodology to evaluate and optimize the energy consumption of the largest Italian WWTP using limited, preliminary energy audit data. An integrated modeling platform linking wastewater treatment processes, energy demand, and production sub-models is developed. The model is calibrated using a stepwise procedure based on available data. Further, a scenario-based optimization approach is proposed to obtain the non-dominated and optimized performance of the WWTP. The results confirmed that up to 5000 MWh annual energy saving in addition to improved effluent quality could be achieved in the studied case through operational changes only.Autosomal recessive cerebellar ataxias (ARCA) are characterized by the abnormal structure of the cerebellum and spinal cord. Spinocerebellar ataxia type 18 (MIM 616204), one of the ARCA, is caused by the loss-of-function mutations of the GRID2 gene due to deletions. Missense mutations in the GRID2 cause ataxia with the gain-of-function mechanism. We report a homozygous GRID2  duplication in childhood-onset ataxia in two siblings. The clinical exome sequencing was performed on one of the siblings. No disease-causing mutations were reported as a result of the clinical exome test. Chromosomal microarray analysis was performed on the entire family using Affymetrix Optima® chips. Chromosomal microarray analysis showed a ~ 121-kb homozygous duplication of GRID2 (arr[GRCh37]4q22.2(94426536_94613158) × 4), including exon 14, in both siblings. Previously, GRID2 has been associated with an autosomal recessive (loss-of-function) and autosomal semi-dominant (gain-of-function) forms of ataxia. To the best of our knowledge, this is the first study to identify a homozygous duplication of GRID2 causing loss of function of the GluRD2 protein. These findings provide us with the conclusion that copy number variation analyses should be in the diagnostic process of autosomal recessive ataxia types.Previous diffusion tensor imaging (DTI) studies have reported that both mild cognitive impairment (MCI) and Alzheimer's disease (AD) revealed microstructural changes [fractional anisotropy (FA)]. However, these results were not conclusive. The purpose of this meta-analysis was to identify the consistent FA alterations and the differences between MCI and AD. Case-control studies investigating MCI and AD using FA were searched in the online databases. The quantitative FA value of cognition-related brain regions was extracted and the standardized mean difference (SMD) with 95% confidence interval (CI) was calculated using fixed or random effect models. Twenty six studies with a total of 1,021 patients were included in this meta-analysis. Significantly decreased FA in patients with AD were identified in the left frontal lobe, corpus callosum (CC), fornix, hippocampus (HP), cingulate gyrus (CG), cingulate bundle (CB), uncinate fasciculus (UF), superior longitudinal fasciculus(SLF), the inferior fronto-occipital fascicles (IFOF), and the inferior longitudinal fasciculus(ILF) relative to MCI in this meta-analysis. Prior uptake of cervical cancer screening was associated with number of live births (p=0.001) and area of residence (p=0.04). And Willingness to screen was significantly associated with age groups (p=0.03) and the number of live births (p=0.03). Moreover, we found that younger age and urban residence was positively associated with awareness and uptake of cervical cancer screening. Willingness was found to decrease as age increased.

The study found that despite older women's higher risk of cervical cancer, those who indicated willingness to screen were younger. Additional education, health promotion, and integration of cervical cancer screening services is needed to improve cervical cancer awareness and screening uptake at the HIV clinic.<br />.

The study was to investigate anti-migration and invasion effects of astaxanthin (ATX), a natural carotenoid derivative distributed in marine environments, against A172 human glioblastoma cells.

Cell viability after ATX treatment was measured by MTT assays. Tumor cell migration and invasion were observed by scratch and Boyden chamber assays, respectively. Expression of MMP-2 and activity of MMP-9 were observed by immunoblotting and gelatin zymography, respectively.

ATX up to 150 µM was not toxic to A172 cells at 48 h post-treatment. In contrast, ATX at 50 and 100 µM significantly decreased migration and invasion of A172 cells at 24 and 48 h post-treatment. Metastatic-reducing effect of ATX is associated with the reduction of MMP-2 and MMP-9 expressions in a dose-dependent manner.

Cervical cancer is preventable. In Malaysia, women are found to have good awareness of the disease and yet, the Pap smear uptake is still poor. Measuring health literacy level could explain this discrepancy. This study aims to determine the relationship between health literacy, level of knowledge of cervical cancer and Pap smear with attitude towards Pap smear among women attending pre-marital course.

A cross sectional study was performed in three randomly selected centres that organised pre-marital courses. All Malay Muslim women participants aged 18 to 40 years old were recruited while non-Malaysian, illiterate, and had hysterectomy were excluded. Validated self-administered questionnaires used were European Health Literacy Questionnaire (HLS-EU-Q16 Malay) and Knowledge and attitude towards Cervical Cancer and Pap Smear Questionnaire. The mean percentage score (mean± SD) was calculated, with higher scores showed better outcomes. Multiple linear regression was used to measure the relationship of independf Pap smear improves the attitude towards Pap smear. Pre-marital course is an opportunistic platform to disseminate information to improve health literacy and knowledge of cervical cancer and Pap smear screening.
We investigated human papillomavirus (HPV) infection and detected anal squamous intraepithelial lesions by modified liquid-based cytology (LBC) and p16/Ki67 dual-staining.

Anal swabs (n=393) were collected from patients with HIV infection. Anal cells were kept in 95% ethyl alcohol for modified LBC. DNA was extracted from cells for HPV detection and genotyping using real-time PCR and reverse line blot hybridization.

Nine samples (2.3%) were unsatisfactory specimens, 74.8% (294/393) were negative for intraepithelial malignancies (NILM) and 22.9% (90/393) exhibited squamous intraepithelial lesions (SIL). In the latter category, 13.7% of samples (54/393) contained atypical squamous cells of undetermined significance (ASCUS), 6.9% (27/393) were classified as low-grade SIL (LSIL) and 2.3% (9/393) as high-grade SIL (HSIL). A total of 331 from 393 swab samples were suitable for detection of HPV infection. Among these, 34.1% (113/331) were positive. HPV 58 (15.9%) was the most common genotype, followed by HPV 18 (14.2%) and HPV 16 (11.5%). The severity of abnormal cells was significantly associated with HPV infection. Dual staining with p16/Ki-67 was performed on 130 samples in 30.8% (40/130) of samples positive staining was significantly associated with severity of abnormal cells. https://www.selleckchem.com/products/Acadesine.html Agreement between cytology, p16/Ki67 dual-staining and high-risk HPV detection was 100% in HSIL samples. The genetic hallmark of CML is known as the appearance of t(9;22)(q34.1;q11.2) (BCR-ABL1) which is present in more than 95% of cases. Here, we demonstrated practical laboratory tools for monitoring of BCR-ABL1 transcripts in chronic myeloid leukemia patients undergoing TK inhibitor therapy.

Real time quantitative PCR and direct sequencing were performed for monitoring of BCR-ABL1 transcripts in 245 treated CML.

At month 3 after first time point of monitoring, we found that 89% (218/245), 2% (5/245), and 9% (22/245) of patients are determined as optimal, warning, and failure response, respectively. The responses to TKI were slightly decreased at months 6 as following 73% optimal (180/245), 18% warning (43/245), and 9% failure response (22/245). Additionally, responses to TKI were gradually decreased at month 12 after first time point of monitoring as following 65% optimal (160/245), 13% warning (31/245), and 22% failure (54/245). We could detect 20% (49/245) of patients positive for BCR-ABL1 TKD mutations.
