Patients with COVID-19 could be vulnerable to other infections owing to multiple comorbidities with severe COVID-19, prolonged hospitalization, and SARS-CoV-2-associated immune dysfunction. These patients have acquired secondary bacterial infections or superinfections, mainly bacteremia and urinary tract infections. This review will summarize the prevalence of A. baumannii coinfection and secondary infection in patients with COVID-19.Ticks and associated tick-borne diseases in livestock remain a major threat to the health of animals and people worldwide. The purpose of this cross-sectional DNA-based study was to assess the presence and prevalence of tick-borne Francisella sp., Rickettsia sp., and piroplasmids in ticks infesting livestock, and to estimate their infection rates. A total of 562 tick samples were collected from camels, cows, sheep, and goats in the Emirates of Abu Dhabi, Dubai, and Sharjah from 24 locations. DNA was extracted from ticks and PCR was conducted. We found that Hyalomma dromedarii ticks collected from camels had Francisella sp. (5.81%) and SFG Rickettsia (1.36%), which was 99% similar to Candidatus Rickettsia andeanae and uncultured Rickettsia sp. In addition, Hyalomma anatolicum ticks collected from cows were found to be positive for Theileria annulata (4.55%), whereas H. anatolicum collected from goats were positive for Theileria ovis (10%). The widespread abundance of Francisella of unknown pathogenicity and the presence of Rickettsia are a matter of concern. The discovery of T. ovis from relatively few samples from goats indicates the overall need for more surveillance. Increasing sampling efforts over a wider geographical range within the UAE could reveal the true extent of tick-borne diseases in livestock. These species were then targeted at six field sites across Florida and sampled from June to September 2020, using drift fence arrays, cover boards and fishing. We captured 125 lizards and collected 233 immature I. scapularis. DNA was extracted from ticks and lizard tissue samples, followed by PCR testing for Borrelia spp. Of the captured lizards, 69/125 were infested with immature I. scapularis. We did not detect Borrelia spp. from tick or lizard tissue samples. Overall, we found that lizards are commonly infested with I. scapularis. However, we did not detect Borrelia burgdorferi sl. The purpose of this cross-sectional DNA-based study was to assess the presence and prevalence of tick-borne Francisella sp., Rickettsia sp., and piroplasmids in ticks infesting livestock, and to estimate their infection rates. A total of 562 tick samples were collected from camels, cows, sheep, and goats in the Emirates of Abu Dhabi, Dubai, and Sharjah from 24 locations. DNA was extracted from ticks and PCR was conducted. We found that Hyalomma dromedarii ticks collected from camels had Francisella sp. (5.81%) and SFG Rickettsia (1.36%), which was 99% similar to Candidatus Rickettsia andeanae and uncultured Rickettsia sp. In addition, Hyalomma anatolicum ticks collected from cows were found to be positive for Theileria annulata (4.55%), whereas H. anatolicum collected from goats were positive for Theileria ovis (10%). The widespread abundance of Francisella of unknown pathogenicity and the presence of Rickettsia are a matter of concern. The discovery of T. ovis from relatively few samples from goats indicates the overall need for more surveillance. However, little is known about the effectiveness of topical tacrolimus in this patient population. The aim of this systematic review was to evaluate the current state of literature to evaluate the safety and effectiveness of rectal administration of topical tacrolimus, in the form of suppository, ointment, and/or enema in patients with ulcerative proctitis, perianal Crohn's disease (CD), and chronic refractory pouchitis. Electronic database searches were conducted in international databases since their inception until February 2020. Study subjects were categorized into three groups topical tacrolimus for patients with proctitis, perianal CD, and chronic refractory pouchitis. The primary end point of this study was the remission rate. Secondary end points were response rate and the incidence of AEs. Eleven studies were included in the final assessment in this systematic review. This provided information from 188 patients. Tacrolimus was administered topically as suppositories, ointment, or enema. Clinical remission was achieved in 57.1%, 57.14%, and 70.0% in patients with proctitis, fistulizing perianal CD, and chronic pouchitis. The most commonly reported side effect was perianal itching and burning. Reversible nephrotoxicity occurred in a single patient. No clear correlation was found between blood levels and clinical outcomes. Topical tacrolimus is effective for a subset of patients with IBD. The adverse effects were minimal and tolerable. Cancer-associated fibroblasts (CAFs) are an important component of the tumor microenvironment, but a systematic investigation of their molecular characteristics and clinical relevance are lacking. Here, we sought to compare CAFs across multiple cancer types to identify critical molecular pathways activated in CAF subtypes, which may contribute to clinical outcome, disease progression, and immunotherapy resistance.

We performed integrated analysis of CAFs from melanoma, head and neck squamous cell carcinoma, and lung cancer, and identified the molecular characteristics that are distinctly active in each CAF subtype. Gene signatures for individual CAF subtypes were identified and used to study the association of subtype abundance with clinical outcome and immunotherapy resistance.

We identified six CAF subtypes (pan-CAF) shared across cancer types and uncovered the molecular characteristics and genetic pathways distinguishing them. Interestingly, these CAF subtypes express distinct immunosuppressive factors, such as CXCL12 and CXLC14, and stem cell-promoting factor IL6. In addition, we identified novel transcriptional drivers (MEF2C, TWIST1, NR1H3, RELB, and FOXM1) key to CAF heterogeneity. Furthermore, we showed that CAF subtypes were associated with different clinical outcomes and uncovered key molecular pathways that could activate or suppress cancer progression or were involved in resistance to anti-PD1 or anti-PD-L1 immunotherapy.

BAY1436032, an inhibitor of mutant isocitrate dehydrogenase 1 (mIDH1), was active against multiple IDH1-R132X solid tumors in preclinical models. This first-in-human study was designed to determine the safety and pharmacokinetics of BAY1436032, and to evaluate its potential pharmacodynamics and antitumor effects.

solid tumors.

In dose escalation, 29 subjects with various tumor types were administered BAY1436032 across five doses (150-1,500 mg twice daily). BAY1432032 exhibited a relatively short half-life. Most evaluable subjects experienced target inhibition as indicated by a median maximal reduction of plasma R-2-hydroxyglutarate levels of 76%. BAY1436032 was well tolerated and an MTD was not identified. A dose of 1,500 mg twice daily was selected for dose expansion, where 52 subjects were treated in cohorts representing four different tumor types [lower grade glioma (LGG), glioblastoma, intrahepatic cholangiocarcinoma, and a basket cohort of other tumor types]. The best clinical outcomes were in subjects with LGG (
= 35), with an objective response rate of 11% (one complete response and three partial responses) and stable disease in 43%. As of August 2020, four of these subjects were in treatment for >2 years and still ongoing. Objective responses were observed only in LGG.

We examined the impact of PNFlx treatment on the formation and maintenance of perineuronal nets (PNNs), extracellular matrix (ECM) structures that deposit primarily around inhibitory interneurons, and mark the closure of critical period plasticity. PNFlx treatment evoked a significant decline in PNN number, with a robust reduction in PNNs deposited around parvalbumin (PV) interneurons, within the CA1 and CA3 hippocampal subfields at postnatal day (P)21 in Sprague Dawley rat pups. While the reduction in CA1 subfield PNN number was still observed in adulthood, we observed no change in colocalization of PV-positive interneurons with PNNs in the hippocampi of adult PNFlx animals. PNFlx treatment did not alter hippocampal PV, calretinin (CalR), or Reelin-positive neuron numbers in PNFlx animals at P21 or in adulthood. We did observe a small, but significant increase in somatostatin (SST)-positive interneurons in the DG subfield of PNFlx-treated animals in adulthood. This was accompanied by altered GABA-A receptor subunit composition, increased dendritic complexity of apical dendrites of CA1 pyramidal neurons, and enhanced neuronal activation revealed by increased c-Fos-positive cell numbers within hippocampi of PNFlx-treated animals in adulthood. These results indicate that PNFlx treatment alters the formation of PNNs within the hippocampus, raising the possibility of a disruption of excitation-inhibition (E/I) balance within this key limbic brain region.The reproducibility and translation of neuroscience research is assumed to be undermined by introducing environmental complexity and heterogeneity. Rearing laboratory animals with minimal (if any) environmental stimulation is thought to control for biological variability but may not adequately test the robustness of our animal models. Standard laboratory housing is associated with reduced demonstrations of species typical behaviors and changes in neurophysiology that may impact the translation of research results. Modest increases in environmental enrichment (EE) mitigate against insults used to induce animal models of disease, directly calling into question the translatability of our work. This may in part underlie the disconnect between preclinical and clinical research findings. Enhancing environmental stimulation for our model organisms promotes ethological natural behaviors but may simultaneously increase phenotypic trait variability. To test this assumption, we conducted a systematic review and evaluated coefficients of variation (CVs) between EE and standard housed mice and rats. Given findings of suboptimal reporting of animal laboratory housing conditions, we also developed a methodological reporting table for enrichment use in neuroscience research. Our data show that animals housed in EE were not more variable than those in standard housing. Therefore, environmental heterogeneity introduced into the laboratory, in the form of enrichment, does not compromise data integrity. Overall, human life is complicated, and by embracing such nuanced complexity into our laboratories, we may paradoxically improve on the rigor and reproducibility of our research.

Saccharomyces pastorianus, immobilized by a simple entrapment technique and by microencapsulation in alginate was characterized using SEM, EDAX, pHPZC and the biosorption behavior toward organic pollutant, such as cationic dye. The biosorption experiments were studied by assessing, in a first stage, the influence of the most important operational physical parameters on the efficiency of the biosorbent the initial concentration of the dye, the contact time between phases, the temperature, the dye solution pH, the biosorbent granule size, and the amount of biosorbent. The highest sorption capacity was obtained for the biosorbent obtained by microencapsulation, at pH 9, at biosorbent dose of 5.28 g/L and a contact time of about 100 min. The biosorption equilibrium was then studied by modeling the data on the Langmuir, Freundlich and Dubinin- Radushkevich isotherms. The Langmuir model is best suited for experimental data on both particle sizes leading to a maximum biosorption capacity of 188.679 mg/g at room temperature. The values of the adsorption energy, E, obtained with the help of the Dubinin-Radushkevich model-suggest that the type of mechanism (physical or chemical) involved in the biosorption process depends on the particle size of the biosorbent. The results confirm that the residual microbial biomass of Saccharomyces pastorianus immobilized in a polymeric matrix such as sodium alginate, can be considered an efficient biosorbent in retaining cationic organic dyes present in aqueous solutions in moderate concentrations.A novel and cost-effective method for the fabrication of highly conductive Al/Ag core-shell structured microspheres was proposed and investigated. The oxidative co-deposition of catechol and polyamine was firstly performed to modify the surface of the aluminum microsphere. Then, a two-step electroless plating was conducted to fabricate the Al/Ag microspheres. During the first step of the electroless plating process, the surface of the aluminum microsphere was deposited with silver nanoparticle seeds using n-octylamine and ethylene glycol. Then, during the second step of the electroless plating process, silver particles grew evenly to form a compact silver shell on the surface of aluminum via a silver mirror reaction. According to the scanning electron microscope and energy dispersive X-ray results, a compact and continuous silver layer was successfully generated on the surface of the aluminum. The valence of the sliver on the surface of the aluminum was confirmed to be zero, based on the X-ray photoelectron spectrometer and X-ray diffractometer analyses. As a result, the as-prepared Al/Ag microspheres exhibited a high conductivity of 10,000 S/cm. The Al/Ag/MVQ composite demonstrated low electrical resistivity of 0.0039 Ω·cm and great electromagnetic interference shielding effectiveness at more than 70 dB against the X-band, and this result suggests that the as-prepared composite is a promising conductive and electromagnetic shielding material.To achieve a preferable compatibility between liquid silicone rubber (LSR) and cable main insulation in a cable accessory, we developed SiC/LSR nanocomposites with a significantly higher conductivity nonlinearity than pure LSR, whilst representing a notable improvement in space charge characteristics. Space charge distributions in polarization/depolarization processes and surface potentials of SiC/LSR composites are analyzed to elucidate the percolation conductance and charge trapping mechanisms accounting for nonlinear conductivity and space charge suppression. It is verified that SiC/LSR composites with SiC content higher than 10 wt% represent an evident nonlinearity of electric conductivity as a function of the electric field strength. Space charge accumulations can be inhibited by filling SiC nanoparticles into LSR, as illustrated in both dielectric polarization and depolarization processes. Energy level and density of shallow traps increase significantly with SiC content, which accounts for expediting carrier hopping transport and surface charge decay. Finite-element multiphysics simulations demonstrate that nonlinear conductivity acquired by 20 wt% SiC/LSR nanocomposite could efficiently homogenize an electric field distributed in high-voltage direct current (HVDC) cable joints. Therefore, in this article, waterborne polyurethane (WPU) as a promising new polymer reinforcement material was proposed to reinforce the road demolition waste (RDW), and the mechanical performance of WPU-reinforced RDW (named PURD) was investigated using triaxial unconsolidated and undrained shear (UU) and Scanning Electron Microscope (SEM) tests. The results showed that under the same curing time and confining pressure, the shear strength of PURD increased with the increase in WPU content. When the WPU content was 6%, the WPU presented the best reinforcement effect on RA. The failure strain of PURD increased with the increase in confining pressure, but increased first and then reduced with the increase in WPU content. The specimens with 5% WPU content showed the best ductility. At the curing time of 7 and 28 days, the internal friction angle and cohesion of PURD increased with the increase in WPU content, and they reached a maximum when the WPU content was 6%. The internal friction angle barely budged, but the cohesion increased obviously. In this review, we summarize recent experimental investigations of the interfacial lubrication properties of surfaces coated with bottlebrush bio-lubricants and bioinspired bottlebrush polymers. We also discuss recent advances in understanding intermolecular synergy in aqueous lubrication including natural and synthetic polymers. Accordingly, the permeability of CH4 in polyvinylidene fluoride (PVDF) containing crude oil was studied at the normal service conditions by molecular simulations. The results showed that the solubility coefficients of CH4 in PVDF containing crude oil were much lower than those in pure PVDF. It can be concluded that the crude oil molecules absorbed into PVDF occupied certain adsorption sites, resulting in a decrease in the adsorption capacity of CH4 molecules in PVDF. The diffusion coefficients of CH4 in oil-containing PVDF were significantly greater than in PVDF. This is because the absorption of oil molecules leads to the volume swelling of PVDF and then increases the free volume for diffusion. The permeation process showed that CH4 molecules were selective-aggregate adsorbed in the region with low potential energy in oil-containing PVDF firstly, and then they vibrated within the holes of PVDF containing oil in most cases and jumped into the neighboring holes at high temperatures and pressures.Molecular dynamics (MD) simulation was used to study the influence of electric field on Glucagon-like Peptide-2 (GLP-2). Different electric field strengths (0 V/nm ≤ E ≤ 1 V/nm) were mainly carried out on GLP-2. The structural changes in GLP-2 were analyzed by the Root Mean Square Deviation (RMSD), Root Mean Square Fluctuation (RMSF), Radius of Gyration (Rg), Solvent Accessible Surface Area (SASA), Secondary Structure and the number of hydrogen bonds. The stable α-helix structure of GLP-2 was unwound and transformed into an unstable Turn and Coil structure since the stability of the GLP-2 protein structure was reduced under the electric field. Our results show that the degree of unwinding of the GLP-2 structure was not linearly related to the electric field intensity. E = 0.5 V/nm was a special point where the degree of unwinding of the GLP-2 structure reached the maximum at this electric field strength. Under a weak electric field, E < 0.5 V/nm, the secondary structure of GLP-2 becomes loose, and the entropy of the chain increases. When E reaches a certain value (E > 0.5 V/nm), the electric force of the charged residues reaches equilibrium, along the z-direction. Considering the confinement of moving along another direction, the residue is less free. Thus, entropy decreases and enthalpy increases, which enhance the interaction of adjacent residues. It is of benefit to recover hydrogen bonds in the middle region of the protein. These investigations, about the effect of an electric field on the structure of GLP-2, can provide some theoretical basis for the biological function of GLP-2 in vivo.Commercial filaments of poly(lactic acid) (PLA) composites with particulate filler, carbon fiber, and copper powder with different contents were fabricated by FDM 3D printing in XZ-direction at bed temperatures of 45 °C and 60 °C. The effects of additives and bed temperatures on layer adhesion, fracture behavior, and mechanical performance of the PLA composites 3D printing were evaluated. Rheological properties informed viscous nature of all filaments and interface bonding in the PLA composites, which improved printability and dimensional stability of the 3D printing. Crystallinity of the PLA composites 3D printing increased with increasing bed temperature resulting in an improvement of storage modulus, tensile, and flexural properties. On the contrary, the ductility of the 3D printing was raised when printed at low bed temperature. Dynamic mechanical properties, the degree of entanglement, the adhesion factor, the effectiveness coefficient, the reinforcing efficiency factor, and the Cole-Cole analysis were used to understand the layer adhesion, and the interfacial interaction of the composites as compared to the compression molded sheets.
